You have to enable the Spam Quarantine first.

On a single box, go to Montior/Spam Quarantine, and enable the quarantine, and set it up for what ever access you want to give users.

If you're using an SMA, and want all of your ESAs to send messages to a Centralized Quarantine, you need to enable it on the SMA, then on each ESA (or just one if they're clustered) you can go to Security Services, and under Centralized Quarantine, enable the Spam Quarantine there.
