I want to change the 'External Number Mask' field under Directory Number Information for several Voicemail Ports in CUCM. After I save and apply these changes in CUCM, will I need to reset the port group in CUC for the changes to be successful?

Correct, since no changes are made on CUC there is nothing to reset there.
I think you'll just need to reset the voice ports in CUCM. But no harm in resetting from CUC as well.
