When ISE does DOT1X Authentication,, it does not appear to be trusted. I want to import an existing wildcard certificate(PEM+KEY), which results in a false report

Import cert chain as individual PEM formatted certs into trust store.  Make sure the system cert used for EAP is associated with this trust chain.  Remember that client must trust ISE cert.  Make sure client trusts the cert or signing cert.
