Is it possible to work if I have a router which has VRF and I connected it in my ASA which only running a single-context mode?

Depends what you mean.
The ASA does not support VRFs as far as I know but if you have an interface on a router in a VRF you can connect to an interface on the ASA if that is what you mean.
The ASA does not need to be aware of the VRF.
Is that what you are asking ?

yes my concern is that my router will be configure with VRF and will be connected to the ASA. Is that possible, how can I do that? Should I configure the ASA in multi-context mode or just single-context will do? thanks

If it is just one interface on the router in a VRF then you can just connect it to the firewall.
If there are multiple VRFs you may need contexts.
Perhaps if you provided some more details as to what you are trying to do we could help more.
