Hi all,
Would like to know if inter-register signaling (such as R2-compelled, R2-non-compelled) necessary for a E1R2 configuration?
I would like to integrate Cisco 2811 with NEC Neax 2000 using E1R2.

It will be necessary, yes. There is information passed with the inter-register signaling that is required for call setup.

Also there's the little problem that the Cisco IOS CLI requires the inter-register signaling type when you define the ds0-group.
Router(config-controller)#ds0-group 1 timeslots 1-15 type r2-digital ?

Do you have any experience if two-stage dialing is supported over E1 R2 interface?

Two stages dialing will work on E1 R2 just like one stage works as well.
