Jumbo frames on 6500-E

I need to enable jumbo-frames on a 6513-E with a Supervisor 2T module

Do I have to enable this globally? Or simply adjust the MTU size on the interface/vlan?

If I do have to enable it globally, what kind of consequences would it have for existing systems on the switch?

On the 6500 series, you can enable it per interface/vlan and there is no need to reboot.
