No.  If you try to configure a 5596 with multiple N55-M160L3-V2 in the configuration tool you will receive an error "Max [1] N55-M160L3 can be selected per chassis."

Why would you want to put two (or more) Layer 3 module into a single Nexus chassis???

Because the Nexus 5596 L2 forwarding rate is line rate, but we should enable Layer 3 function, if installed one Layer 3 module only, the Layer 3 forwarding rate is 160Gbps only.
