I've created a new video screen layout for our SX80 with the use of TC Console.
The layout was correctly created and uploaded to the SX80, and I can select it from the touch panel.
But when I want to force this layout from the webpage I cannot find it in the list (see attached screenshot)..
With Xconfiguration I do not see the layout, with Xstatus I can find it. Looks like it is volatile.
Did I forget something to make this available for permanent configuration?

Take a look at the SX80 API Guide, pg 135, at "xCommand Video Layout LoadDb". It might get you what you want, other than that, I'm not sure how to get the TC Console layouts to appear within the web interface.

Will this command make the customer layout the default one and avoid the user can select the default layouts from the touch panel?
I see there are 2 option in the command:
Custom: The system uses the custom video layout database which generated by the Cisco TC Console tool.
AutoCustom: The system uses the auto mode part of the custom video layout database.
Not sure what's the difference of these 2 options.

If you want your custom TC Console layout to be the default, you should be able to set it that way from within TC Console. That's what we did for all ours.

The custom TC Console layouts do not appear in the browser GUI (that's a fixed, pre-defined list). They will be selectable via the remote or touch panel on the device though, or via the API.
