I'm planning to implement VPN.Do I still need to buy user licence for my mobile clients using laptop travelling across the globe connecting back to my PIX vpn server.Or I just need to use the Cisco VPN client4.0

Buy a PIX based on your user count - there is no per user licensing of software, the only limit is possible concurrent per user limitations of various pix models. With the 6.3 PIX os, a pix 501 now supports 10 vpn clients (it used to be 5), I think the 506 has a limit as well. I think the 506 has a user limit as well.

If let say I'm doing a site to site VPN,will it mean the site to site connection is calculate under 1 connection?For example for as you said501 support 5 vpn clients?

Yup, the vpn count is the # of tunnels that terminate. SO a 501 with 6.3 could act as a hub for 10 branch offices, or 10 concurrent end users.
