Cisco ESA c190 + ESA PXE Encryption License + Cisco Email Security Inbound  for 2000 users,will this solution works without purchase ESP license ?

ESP is the premium bundle which includes inbound and outbound scanning features.

Your description above shows only inbound scanning features so would be missing DLP. The solution would certainly work if you do not require DLP.
