We configured SPAN on a Cisco switch 3750. After configuration of SPAN, the machine which is connected with Gi3/0/4 is not able to ping source port(Gi3/0/2) IP. Below snapshot is from my Cisco switch

SPAN ports only mirror data that you are sniffing with your analyzer. Once you configure 3/0/4 as the span destination and default the interface it will go into 'monitoring'. Therefore will not be able to ping your source interface.

Thanks for your response.
So my understanding is that, packets will be mirrored from source to destination but we cant ping both interfaces. right ?

Yes. However, your source interface should still be pingable from other interfaces, excluding your destination monitoring port.
