According to the documentation you have to use the following query to see if a user is member of a certain group:
(&(memberOf={g})(proxyAddresses=smtp:{a}))
I searched the AD tree (even with Apache Directory Studio), but I cannot find the 'memberof' object.

Do I have to activate it or did I miss something?

That's right. I'd recommend using Softerra LDAP Browser:

There is nothing to activate, just make sure you are succesfully querying AD.

Setup the LDAP profile for one of your domain controllers, than you should be able to navigate to a user through the LDAP browser and see all the groups they are a member of:
