i got 6 public ip address from my service provider, I have one webserver on the dmz zone which will be hosting the website and mail server on the inside network. The webserver will talk to mail server as the organisation plans to provide web based email access to their employees.

do i need to create access list for webserver to allow the traffic access the mail server

If you want the webserver to be able to talk to the mail server, you have to configure an access list (or old style "conduit permit") permiting traffic from the IP of the webserver to the IP address in which you translated the internal address of the mail server.
