A client would like to migrate off PPTP to IPSec using an RV345 and the 3rd party VPN client option. Does anyone know if there are any docs or instructions out there that describe how to enable this using the native Windows 10 L2TP/IPSec client? I'm finding the internal RV345 documentation sparse.

Hi,
L2TP configuration for Windows 10 would be very similar to what is described in the guide below, just changing PPTP for L2TP.
If you wish to use a 3rd party client for IPsec VPN you can take a look at ShrewSoft or TheGreenBow.

Do you know is it possible to get the OpenVPN Client to work with the Cisco?

RV340 and RV345 have problems with IPsec vpn if you don't work with Cisco AnyConnect. I spent two days talking with Cisco support and they couldn't solve the problem and tall me that there are two options: connect through PPTP (dangerous) or through SSL with AnyConnet (payment software). Cisco support had been not able to configure Shrew and RV340 to establish vpn but this tutorial do it. In my case I needed to activate NEM and disable Split Tunnel.
