Q1. Can we use a single port in different VLAN ?

Q1. Yes, to use a port in a different VLAN, simply reassign it to the new VLAN. Remember a port can belong to only one VLAN at a time.
Q2. You can configure your router using Cisco Configmaker (http://www.cisco.com/warp/public/cc/pd/nemnsw/cm/index.shtml) otherwise here is a brief tutorial on how to configure a Cisco router - http://www.swcp.com/~jgentry/topo/cisco.htm. The relevant IOS command set for your router can be found here http://www.cisco.com/univercd/home/home.htm.
