I have several 1601's. On one of them, Hyperterminal is non-responsive when running under Windows 98, but fine under Windows 2000. On another one, Hyperterminal displays router status messages, but doesn't respond to keyboard input. On yet another one, Hyperterminal doesn't display anything (other than occasional garbage during a reload) nor does it respond to keyboard input. The basic settings on Hyperterminal are correct (9600/8/n/1/n). Any input will be greatly appreciated.

There are issues with Windows 2000 and specific serial port hardware.
Are you running Win2k on a Dell laptop?
On the same exact hardware has 98 worked but Win2k not worked?
Make sure that Hyperterminal is up and running _before_ plugging into the console port.
Make sure you unplug the cable _before_ stopping Hyperterminal.

I have a Windows 2000 machine running on a Dell C600. I've had a Cat3548 reboot on me when running Smarterm Emulation. Does this happen only with Dell computers or do other brands (IBM, Sony, or Gateway) also have the same problem?

You can try also with the Hyperterminal Private Edition on http://www.hilgraeve.com/htpe/index.html, it has many enhancements and is W2K compatible.
