I have a 3DES VPN tunnel running between a 3640 and a 2600 router. I am looking to install VPN modlues in each router to increase the throughput.

Do these modules require any additional configuration in the router?

No configuration is necessary, the router will detect their presence and start using them. You may require a software upgrade so that the software will recognise them, so check with your reseller as to what version is required for the particular module you're installing.
