I have marked some traffic as DSCP value EF (46), and changed the cos-dscp map so 46 map to CoS 5, and I enabled qos on the switch. Is this enough if I want the traffic marked uses Priority Queues into the switch or I have to enable the priority Queue in some way?

Hi
Which switch are you using. You usually have to map the DSCP value to a queue and then mark that queue as the priority queue.
Could you send details of switch and config you have so far.

A Catalyst 6000, I mark the traffic with an ACL and set the DSCP value

I think you have to turn on the priority queue on the port. In IOS the command is: priority-queue out but I don't the how it looks in CatOS.
You can check the port capabilities (show port capabilities) to make sure how it is configured.
If you see something like:
QOS scheduling rx-(1q4t),tx-(2q2t)
Than the priority queue is not turned on.
