If I am creating an EtherChannel between the access switch and the Multilayer switch, does the mode needs to be the same?

- No , but you may leave one end in mode desirable (for 'parent') and the other default (should work) , if not check this document for further options : https://www.cisco.com/c/en/us/td/docs/switches/lan/catalyst4500/12-2/31sga/configuration/guide/config/channel.pdf mode on , usually applies to different vendor solutions.
