How i can stop pinging particular computers from different networks even routing is configured properly and without using acl?

You could choose not to give the clients a default gateway, they will not be able to access anything off their own subnet.
Perhaps use a firewall between subnets but effectively that is just a more advanced ACL as such.
What are you trying to achieve and more importantly. why?

You cannot, you need to use an extended ACL between the IP subnets to stop ICMP traffic.
without using an ACL on multilayer switches or routers all traffic is allowed between subnets.
There are differences for firewalls that using trust zones and levels of security have a different approach.
