Hi Arif,
ISA server are build in firewall by default, I have seen the topology with ISA having two nic one with 10.10.x.x ip and other with 192.168.2.x but it is connected to core switch 192.168.100.x how is the connectivity is there and what is the routing configuration for 192.168.100.x in ISA server i mean reverse route where it is pointing out.
And for icmp allow by default ISA block you need to enable by deploying the policy check out the below link for more information on icmp enablement in ISA.
Hope to Help !!
