This server is controlling  internet for all Vlan's , my question ,

Why all Vlan's can get internet with that it is in switch port mode ?

As Divin suggested,

Can you share the results of the following

From the PC

ping 192.168.2.250
ping 192.168.100.253

Hi Arif,

ISA server are build in firewall by default, I have seen the topology with ISA having two nic one with 10.10.x.x ip and other with 192.168.2.x but it is connected to core switch 192.168.100.x how is the connectivity is there and what is the routing configuration for 192.168.100.x in ISA server i mean reverse route where it is pointing out.

And for icmp allow by default ISA block you need to enable by deploying the policy check out the below link for more information on icmp enablement in ISA.
