Does anyone know the differences between the releases? And why is there a NOTE in the release notes for 6.0.132 that states: "Note - Any release posted after 5.2.130.0 will not be eligible for upgrade to release 6.0.132.0."

The reason for this is:
5.2.148 (or any version after 5.2.130) are released AFTER 6.0.132.0. Hence if you go to 5.2.1480.0 now, you will have to wait the next release of 6.0 that will support the newer database upgrade that 5.2.148.0 does have.

I am running 5.2.130, so I should go to 6.0.132 directly, skipping 5.2.148? Then I should be good going forward from there? So, the significant difference is a new database structure? Are there any other things that I should be aware of going from 5.2.130 to 6.0.132? Any "special" considerations?

I would suggest you to go to 6.0 directly instead of staying on 5.2. In any new release of WCS, there is a change in the db schema hence the upgrade path is sometimes a little tricky as there are maintenance releases of WCS on previous versions released after a next major version.
I suggest you to go through the release notes :
