how can i configure ISR4321 Router with USB to RJ45 console cable. cant see any RJ45 Console port at back of the router.

ISR4321 router have two (2) Console ports:

Maybe do not confuse with @Leo Laohoo comment with numbers 1 and 2 - that is option (in the picture port reference numbers are different- hope you not)

The Cisco datasheet states that the 4321 ISR provides these console ports and Auxiliary port:

• One USB mini-Type B console (placed next to the RJ-45 console port)
• One RJ-45 console port
• One RJ-45 auxiliary port

Provided is an image showing a console cable plugged into the RJ45 console port. Viewing from the back, It is to the right of the USB mini-Type B console port and underneath the RJ-45 AUXiliary port.
I agree that it is not clearly marked...it can easily be mistaken as a second RJ-45 auxiliary port.
