Brand new 2.4.1 agent VM. Status in CX Cloud portal is running. I added our Catalyst Center to the agent. After 2 hours Catalyst Center status remains "Unknown". When running cli commands debug-cxcli agent checkControllerReachability or cxcli agent modifyController I get the error "The service is not up and running, please retry after sometime". I've rebooted the agent VM as well. -Update it has now been 8 hours and no change.

Recommend you open a CX Cloud Support case. You can do that from the Help menu in CX Cloud and select "CX Cloud Support".
