I am having 2921 router. I need q-in-q tunneling. Does WIC-4ESW card support q-in-q?

The WIC-4ESW doesn't, but the HWIC-4ESW and the EHWIC-4ESG do.

You can also do Q-in-Q tunnelling on the router's ports with the "second-dot1q" parameter of the "encapsulation dot1q" subinterface configuration command,

Yes, but you set it up using subinterfaces and the"second dot1q" parameter on the "encapsulation dot1q" configuration command.
