I have Cisco ASA firewall. I want my clients to access my server via RDP via ASA firewall.
There is no direct communication from clients to the server segment
Client -> ASA1 -> ASA 2 -> Server (RDP)
(Source NAT) (Destination NAT)
Both source & destination needs to be NAT for RDP access. The source & destination NAT occurs at different ASA firewalls.
Would like to check whether Microsoft RDP will work in this scenario.
Also please let me know whether Microsoft RDP use TCP or UDP

Source and destination nat can be done on different different ASA so no issue it will work.
TCP port 3389 for RDP access.

Is there any documentation which says it will use TCP 3389
Cause in internet it says it will use both TCP & UDP

By default Microsoft window support both TCP and UDP 3389. It depends on server-side setting what they allowed and client-side setting how they initiate the connection. You can check for Microsoft forum for that.
