I'm on an iMac, Yosemite 10.10.5
My OpenDNS was working a couple weeks ago, and I'm not sure when it stopped. I double checked my Network connection on my computer (Ethernet settings under Network), as well as going to my router address and double checking the settings there, and everything still has the two Open DNS addresses set up as the Primary and Secondary DNS addresses. I cleared/flushed cache, both in Chrome and on my Mac, and still it's not working. I'm not sure what else to try -- everything shows the OpenDNS addresses in the right places, I've restarted computer, Chrome, and the router, all multiple times and it's still not working. Not sure what I'm missing or what else to try.

It sounds like everything you checked was only on your network. Generally speaking if OpenDNS "suddenly" stops working it's because your IP address has changed and wasn't updated for your network. Check to see if your current IP addressed is registered on your dashboard. If it is, proceed with the diagnostic commands rotblitz asked for.
If it isn't, update it, flush your caches and try again. Also, make sure that you have some sort of updater running somewhere on your network.
