I have a CTI port of 1000, and a Softphone of 1001.
When I used Softphone 1001 to call 1000, I got busy signal.
Is there a way I can use JTAPI to open the CTI port 1000. So, CTI port will receive the call from Softphone, and then send a wave file to Softphone ???

You get busy bcuz the CTI port is not registered to any application.
To get a wave file played when you call from Softphone, either you have to "call forward all" the CTI Port 1000 to maybe another phone which is registered to callmanager and pick the call there;
or "call forward all" the cti port 1000 to Unity pilot;
or use another softphone on cti port 1000;
or if you have an TAPI or JTAPI application, like IVR, use the CTI port on that application.
