i'd check disk space on unity and if the message store is mounted, if both good then permissions wizard and MSCW -sync

Ok, here a step-by-step guide on how to check:
1) Go into START > Programs > Microsoft Exchange > System Manager
2) Expand the tree as follows, Administrative Groups > First Administrative Group > Servers
3) Expand the server which Unity is integrated with
4) Open up the First Storage Group and you will see all the mailstores. Check they are all mounted.
You can check all the Exchange servers and stores quickly if you're not sure. More information can be found here:

You might also want to check whether the Exchange server is actually up and running (have you tried pinging it from Unity)?
