Is it possible to backup a serial interface using an ISDN BRI to dial location 1 and in case of failure/unavailability of that, dial to location 2.

This is possible , you just need to add an additional dialer string to the alternate location see below
interface Dialer1
ip address x.x.x.x
encapsulation ppp
dialer in-band
dialer idle-timeout 300
dialer string xxxxxxxxx
dialer hold-queue 100
dialer-group 1
ppp authentication chap
ppp multilink
