Suppose I'm a Tier 3 provider with my own autonomous system number and ip address range, who purchases a transit agreement with a single Tier 1 provider. Is this agreement sufficient to send and receive data from all other users connected to the internet? Or are further agreements with providers further downstream again required?

what kind of Agreement we are considering here, you need to follow the reulations if you looking to alloocate IP address to customer.
for peering that should be ok for your to start.

from a technical point of view it could be enough.
However, to obtain your BGP AS number you must be multi homed ( at least so it was before 4 bytes AS number introduction)
So I would say you need at least two upstream providers to fullfill all requirements ( including fault tolerance).
And what is more important you pay for traffic sent to the upstream provider so most ISP attempt to build as many peering relationships with other entitiies to minimize traffic volume on uplinks. In a peering relationship you don't pay for traffic volume you pay just the operational costs.
Going to an internet exchange point IXC and peering with who is attached there can be a good solution to provide cheaper connectivity.

Probably not ideal to get transit from one single Tier 1 for redundancy purposes, but technically it should provide reach to the entire Internet.
Just wanted to note that there was a major dispute between Cogent and Hurricane Electric, that literally partitioned the IPv6 Internet for the longest time. I am not sure where it stands at the moment. Always good to be aware of these things.
