I would like to connect a Cisco2621 with a Nortel ASN Router over a V.35 serial line. Are there parameters on the Cisco or the Nortel side that I have to configure for a good functionality.

I have not done this so can not speak from any personal experience. But the parameter that comes to mind first is the encapsulation. The default encapsulation on Cisco is HDLC and their implementation is somewhat proprietary. It may interoperate with other vendors but might not. You would be much safer to specify PPP (or something like Frame Relay).

In order for the two beasts to talk to each other, you need to set them both for PPP encapsulation. All other parameters are fine...
