speed command

I have three switches, one is 3850, the other is 4500, the last is nexus 6004,

I want to add speed 1000, duplex full under interface, do I need to reload to take effect for those three switches?

No, you don't need to reload any of the switches although the interfaces may bounce when you modify the settings.

Jon already answered you original question but I want to add current recommendations are to use auto/auto - only if there some problem with that should you need to set speed/duplex.  Also note, when you disable auto/auto, on some devices you also disable auto-MDI and I also believe for copper Ethernet you're effectively disabling some error detection features too.

The 6004 does not come with any copper ports.

To the OP,

On the 6004, you need the 20 Port 1/10gig blade to do this, as the other blades are 10/40 and 100Gig.

You're also not considering FEXs?

I noted that because I thought you are referring to copper ports in your first post. I think (not 100% sure) the OP is taking about fiber ports. If the OP is referring to copper ports, this is not possible, as the 6004 does not come with copper blade.

Ah, yes I was principally thinking about copper ports as for fiber ports speed isn't usually variable, nor is usually duplex for them.

For the 6004 (I have a pair in production), yup the base unit doesn't provide copper ports, but you can set speed/duplex on copper FEX ports.  (On mine, we also have just speed set on QSFP ports.)
