Can you upgrade from TMS 14.3 to 14.6.1 directly or do you have to upgrade each release from 14.3 to 14.6.1

You can upgrade directly to 14.6 without issue, just be sure to check the release notes of each version up to the one you're wanting to go to so you'll understand what changes there have been made if any, also so you're not caught by surprise.

The answer if a direct upgrade can be done is not wrong, but it might need some checks first!

there might be a reason why there is one called:
Cisco TelePresence Management Suite Installation and Upgrade Guide (14.6)
If you look at this guide you will see that some requirements have changed and that there are also other comments regards some deployment or integration types.
Besides that: ALWAYS do backups on regular bases and before every upgrade.
TMS is quite easy to recover from a SQL backup, but that requires that you have one :-)
