I have setup an alternate greeting stating to the caller to please hold while we transfer your call. I wanted to see how I can set this call to forward to an extension (CTI Route Point) in CUCM without any caller input. From the greetings page of the system call handler I dont have the same options as I do when going to the caller input section.

Make a Call Handler that has the extension you want to transfer to in CUCM. Set the Transfer Rules on that Call Handler to always transfer the call instead of sending to greeting. Point your menu option, or your original Call Handler, at the Call Handler that is set to always transfer back to CUCM.
