Is possible via a Call Handler in Unity to flag a voice message as urgent?

Yes, you can turn this on. Look on the "Messages" page for a call handler in the SA and you'll see a section for "After Message Action" - one of the options is to always mark messages to this handler urger, never mark them urgent or ask the caller if they'd like to mark them urgent.
