We've got Youtube blocked via our Ironpoort WSA S170 via the Web Security Manager --> Access Policies, this works fine, however, now we want one video to be available (our director speaking at a conference I believe) but the rest of Youtube blocked. Has anyone attempted this and or sorted it out?

You can try the following workaround:
-- How can I allow a specific YouTube channel, but block all other streaming media?
In order to accomplish this, you will need to set up the following. This article assumes that the Streaming Media category is set to Block in the Access Policies.
Go to Web Security Manager > Custom URL Category and create a new category labeled 'YouTube'
Add the following entries to the Sites field:
c.youtube.com, .c.youtube.com, ytimg.com, .ytimg.com
Add the following entries to the Regular Expressions:
www\.youtube\.com/.*<ChannelName>
youtube\.com/videoplayback
www\.youtube\.com/crossdomain\.xml
youtube\.com/generate
Go to Web Security Manager > Access Policies and create a new policy.
Choose an Identity to apply to the new policy or select the 'YouTube' custom URL category as a membership criteria.
Edit the URL Categories for the new policy and set the action for the YouTube custom URL category to either Monitor or Allow.
Submit and commit the changes.
