Is there a way for a Cisco spark board to make a call to Skype for Business client and vice versa, or whether there is interoperability between Spark and S4B and if so, how it is implemented.

There is no direct interop. You can call into Cisco WebEx CMR and do it that way but that’s just video. White boarding is exclusive to Spark service

Is this still the case? I have a customer on non-Cisco phone system that isn't ready to change just yet, but wants to implement Spark Boards and Room Kits registered to Spark. They use S4B today and want to be able to call directly to and from S4B and Spark endpoints.
They were not able to call the Spark Board directly, but were able to call into a WebEx using the @lync.webex.com address. Is this still the only way for S4B and Spark registered endpoints to make calls?

Yes correct WebEx is still the "transcoding" engine to conver SVC to AVC and RDP to BFCP. Or use CMS as an another option. Direct calling from Spark which is a SIP registration service to SFB without that mixing in the middle isn't possible
