when Assistant uses their own handset to answer the shareline of Manager, they are then unable to transfer the call to the Manager as the Manager's line is in use.

Hi there,
In your example, what you are seeing is the expected behavior as you can't transfer to the same DN/Extension that you are already engaged on. This is where most people use an Intercom setup so that when the Assistant answers the "shared line" call they can put it on hold and contact the Manager to let them know they have a new call and provide the appropriate details.
