A reboot is not required. Depending on your forest design though, you may need to wait for replication to occur before seeing the new objects from your domain. For example, we performed the schema updates on our forest root. But we are one of 9 domains in the forest. Once replication occurred, give or take 15 minutes, we were able to see the updated/new objects from our domain, which is where our Unity is configured.
