Assuming there are two distribution switches (6509) and they each have vlan 2,3,4,5 and interface vlan 2,3,4,5 configured for access layer devices. I'm configuring HSRP active by alternating between two switches for odd and even numbered vlans. Supposedly to have load balancing. In this case, should I also configure spanning-tree vlan priority for those vlans to be consistent with the HSRP config or it doesn't matter at all? I'm looking it more from a load balancing perspective.

Yes you should otherwise you will find that your interconnect link between your 2 distribution switches will have to carry more traffic.
Does it really matter - not if your interconnect has enough bandwidth but you may as well.

Just configure to have your root primary on the hsrp active side for each .

Do you mean if I make switch 1 the HSRP active for vlan 10, also make switch 1 lower spanning tree priority for vlan 10? and use switch 2 the same way for vlan 11, and so on?

Yes, that is exactly what you should do. Match the spanning-tree root to the HSRP active gateway per vlan.
