We have MR42's where the encryption mode is WPA2 and we would like to change it to WPA2 and WPA3 (transition mode). In doing this, will it have users re-authenticate and require them to log in to the network again?

The users will disconnect from the WiFi for a brief second and automatically reconnect back.
They should not be prompted to enter their passwords, assuming they saved the password in their device OS.

Just a note on my experience - I have had nothing but trouble using WPA3 transitional mode. There are very few WPA3 clients, but plenty of WPA2 clients that fail in different ways when they see WPA3 advertised as an option.
So if you enable it be prepared for devices that just won't be able to connect, or can connect but have reliability issues.
