I need to change the mobility group on 12 controllers, my question is will the AP's or controllers need to reboot after these changes are made? What tyoe of service interuption would this change cause?

You can change them without a reboot but your mobility will be down. That's really all the impact.

Roaming between WLCs and guest anchor will get distrupted until the tunnel comes back up.

What about any stations that are currently connected after a layer 3 roam? Given that the roam is established to the second controller dependent on the mobility group, would changing the group name drop the station? Just thinking out loud as usually I would ask for a maintenance window for a change like this and not worry about service impact.

I agree I would ask for a maintance window as well. Mob groups are pretty important and you could hang up a client in a L3 roam.
