CUCM configuration for Intercom

If we configure three CTI route point in CUCM and each will be mapped with distinct DN and we call from our application to any DN  (where CLI/ Caller will be physical device number)  then should call will land on respective CTI route point?

Also, how call can be auto accepted on CTI route point?

CTI Route Points cannot connect a call on their own. They are effectively a load balancer: your application would need to redirect the call to an available CTI Port for if for it to actually connect and send/receive audio.
