It depends on the whole setup and the most frequent pattern of traffic. But I think per destination or Source MAC address will work fine.

what about the src-dst-ip???

I don't think there is difference between src-dst-ip or src-dst-mac in terms of functionality, either of them is a good one to use but again it depends on your network setup.I believe the defualt is src-dst-mac

Whichever one gives the best number of changing values for your traffic's flows.  Often src-dst-IP works well in most cases as it handles traffic to/from hosts on the same L2 network (except between the same two hosts, for those port number help), it handles traffic to/from a host on the L2 network, and it handles traffic that transits the L2 network (e.g. router to router).  An even better mix is 6500 VSS's src-dst-mixed-ip-port.
