Hello:
In the following configuration
Access-list 300 permit ip 192.168.1.0 0.0.0.255 172.16.0.0 0.0.0.255
match ip address 300
set interface Gi3/48
interface vlan 2
ip address 192.168.1.254 255.255.255.0
ip policy route-map MAIN
Does the interface Gi3/48 in the set interface option have to be a L3 interfaces? I mean, does it have to have and Ip address?

I tried it on my lab switch and it doesnot throw me an error. I beleive that it should work but i cannot push the traffic across it to make sure its working.

What platform are you configuring this on? I believe it needs to be L3, but it may depend on platform.
For instance if this is a 6500 you can see what is supported here in the config guide:
