I Must power off core switch 4510R+E when replace Supervisor slot ? or its normal during system on ?

If the chassis only have a single supervisor card, powering down is necessary.
If the chassis is dual-supervisor card, then it is not necessary. Just make sure the new/replacement supervisor card is running the same IOS version as the active one.
IMPORTANT: Whether it is a single or dual-supervisor card, it is good practice to connect a console cable to the to the new/replacement line card and record the boot-up process in case troubleshooting is required.
