Can I use twinax cable between two nexus 5596 switches for setting up the peer-link? Also is it possible to use the twinax cable between 5596up and N2K-C2348UPQ4F?

Yes, you can use twinax for the peer link between 2 5596 switches. As for between 5596 and FEX, I have never used twinax, but usually when you purchase the FEX, it comes with bidi optic of FET for connectivity to the 5596.

Do we have to use FET when connecting FEX with N5K or N7K? or can we use 10gb-sr sfp too and then normal fiber cable

Yes you can use normal 10gbsr optics if you want. The FET optics are cheaper though.
