Is it mandatory that, first we have to do audio call for making video call or we can have direct video call ?

It would help to know what equipment/devices you are dealing with or thinking about.

H.323 or SIP video calls include both audio and video channels. The protocols for audio and video are negotiated during call setup.

Actually i am using virtual CUCM and have configured two soft phone on it. i am first making audio call and convert it into video. so just wondering that, Is it possible for direct video call without making first audio call.

Which soft-phone you use?

You may use cisco jabber for audio and video calls. You can disable/enabled video in device capability.
